Influence of lipid rafts on pattern formation during T-cell adhesion

چکیده
Abstract Adhesion of T cells to antigen presenting is mediated by the TCR–MHCp and LFA1–ICAM1 protein complexes. These intercellular complexes segregate form characteristic special patterns in cell contact zone. Previous studies have attempted explain mechanisms formation these patterns. While emphasis has been put on membrane elasticity active cytoskeletal transport, it remains unclear whether how pattern process related lipid rafts, which are nanoscale molecular clusters enriched cholesterol saturated phospholipids membranes. Using Monte Carlo simulations a statistical mechanical model for T-cell adhesion, we find that rafts can lead intermediate with ring around central domain even absence transport receptor (TCR) molecules toward center In presence TCR accelerate this monocentric pattern. We also strong stabilizing effect after removal transport. Our results not only help recent experimental observations, but demonstrate cooperate during immunological synapse formation.

Pattern formation during T-cell adhesion.
T cells form intriguing patterns during adhesion to antigen-presenting cells. The patterns are composed of two types of domains, which either contain short TCR/MHCp receptor-ligand complexes or the longer LFA-1/ICAM-1 complexes. The final pattern consists of a central TCR/MHCp domain surrounded by a ring-shaped LFA-1/ICAM-1 domain, whereas the characteristic pattern formed at intermediate times...

متن کاملHave we become overly reliant on lipid rafts? Talking Point on the involvement of lipid rafts in T-cell activation.
During the past decade, the lipid-raft hypothesis has focused attention on the role of membrane domains in controlling cellular functions. Among the best-studied roles of lipid rafts is the regulation of T-cell signalling. In particular, a model has emerged in which lipid rafts regulate protein-protein interactions during signalling in a cholesterol-dependent manner. Does this model provide the...
متن کاملThe inner side of T cell lipid rafts.
A key question in understanding the functional role of lipid rafts is whether lipid microdomains at the plasma membrane outer leaflet are coupled to lipid microdomains at the inner leaflet. By using a cyan-fluorescent protein (CFP) targeted to inner plasma membrane rafts of Jurkat T cells, we found that raft domains at the outer and inner leaflets are physically coupled and that this coupling r...
متن کاملT-cell antigen receptor triggering and lipid rafts: a matter of space and time scales. Talking Point on the involvement of lipid rafts in T-cell activation.
T-cell antigen receptor triggering mechanisms and lipid rafts are of broad interest, but are also controversial topics. Here, we review some recent progress in these two research fields, which has been accomplished mostly in live cells and with the use of advanced technologies. We then discuss the potential relationship between membrane-domain organization and T-cell antigen receptor-triggering...
